This one's been in my feed since yesterday, a MutaT7-based continuous evolution system for transcription factors in E. coli. Most in vivo continuous evolution setups couple mutation to phage infectivity (ie PACE) or mutate an entire plasmid (T7 ORACLE). MutaT7 systems allow for targeted gene mutation, and this work by Zhang and colleagues extends it to evolving transcription factors using a toxin-antitoxin system. One neat example is they flipped a transcription factor's regulatory logic, evolving PsiR from a d-allulose-activated switch into one that's repressed by d-fructose/d-mannose instead.
