I think it works on two levels. On one level, it's Curry laughing at how funny the line is.
On a different level, the character themselves are giving a shit-eating grin, in their insanity thinking that they have the upper hand, and smirking at their own ingenuity.
The scene just works really well and the execution is amazing.

Apparently Toad had more lines int he original script and kept asking rhetorical questions about Toads. They we all cut but they kept Storm’s line here in, but without the background lines it comes off as so cheesy and weird.

Is his character a bad liar? So they did intend on killing that old lady?
Is his character telling the truth, and Marky Mark's acting is about as convincing and sincere as a child whose been caught with their hand in the cookie jar?

As much as this line is memed, it kinda made sense in context: dude became overly reliant on a magical artifacts that healed him even after he recived wounds that should have been fatal
This line is said as a "it's dumbly obvious: people die when they are killed, i should really stop assuming that i will be magically saved again"

Yeah, Fate is full of quotes that are memed around but more often than not makes sense in context, some for translation issues
"The archer class is really made of archers" (Rin making this comment when Archer proved his skills, the first "archer" is english, the second is in japanese, so the english screenshot looks weird)
And some with a bit more background "so you dodged my undodgeable blow, saber" (Gae Bolg can't be dodged, but high magic resistance and luck stats from Saber allowed her to be hit, but with a non-lethal blow)

Iirc In cyberpunk, a doll chip allows a persons personality to be entirely overridden.
It's used in escorts work cause clients will have a perfect partner for their wants, etc.
In this case they used it to upload a bunch of combat knowledge to the dolls so they could fight gangsters.
Maiko has one as well and says that just before going hostile in one path for the quest, typically being so weak all V's (the pc) kill her immediately.

Which is funny because IIRC one of the other characters with a doll chip demonstrates how powerful it is by sparring with V,
and if you don't have a good build it's a tough fight, and even if you win (again, IIRC)V admits it was a tough fight.But Maeko apparently fitted herself with the budget version or forgot to switch it on or something, because you can literally take her down like any other gonk, it's hilariously easy. Which makes the line even funnier on replays.

Dolls are hookers in Cyberpunk 2077, they have this chip that activates "doll mode", it's like a role play character that takes over their body, does the deed and then they're back to normal, not having experienced any of it. Some of them have a modified doll chip that's programmed to kill, it was a whole quest to figure out what was happening then you run into this bitch who threatens you once you figure it out.
