Advice First time applying
I’ve been told for a while I should be a voice actor and I’m about to apply for every male character character to boost my chances while showing them what I’m made of, I’m just wondering if ya’ll got any tips for the 16 bar cut of a musical theatre songs I have to do and how to keep the voice going strong while singing? Especially when it’s a deep rough voice I’m trying to do?

Well how deep are you trying to go? Bass? Then check out hey little songbird from hadestown. Baritone? While Gleb on the Anastasia musical is a tenor it's supposed to be a baritone role normally so look into Still/Neva Flows.
Now make sure you're warming up. Search up some exercises to warm up your vocal cords, if not you can strain and hurt your voice. When you're singing especially in a deeper register you're going to want to dip your chin down and close up your mouth. Don't so that. Try to make sure you're keep your mouth open and your chin up. When you raise your eyes you might feel the back of your throat raise, that's your soft palette and keeping that up while singing can help.
Now breathing. You're going to want to really use your diagphram. You'll really feel it when your stomach extends. Use this air to push your lyrics out.

Well the thing is the voices aren’t specified, so I think I have to come up with what I think they’d sound like and so far I’ve came up with one that’s good and I have a slight idea for another but the other’s I haven’t a clue but so far the 2 would be deep ones like a deep Yorkshire type accent at least I think it’s Yorkshire

Is this for The Last Fire Sprite? Not every day I find an animated show that has musical elements, let alone being under SAG AFTRA.
There's no quick trick to singing well. It takes practice and guidance. That said, it will definitely help you in acting and other vocal performances. It would not be wasted time as far as furthering a career.
Learn to sing first, then practice applying a character voice while singing. Sounding like the character won't help if the vocal technique isn't there.
If you legitimately don't have the singing down, this might not be the casting call for you. By all means take a shot for the practice. But at the end of the day they're looking for someone who can deliver pro results on both acting and song.

I drink Big Red soda to coat my throat during recordings. Not the healthiest but it works.

Needing 16 bars of a MT song is highly unusual for a VO audition. Is it for a particular casting call?
Also if your voice isn't naturally low and gravely, don't force it. Taking singing lessons and working with a VO coach will help you expand your range healthily. And once you've leveled up thar skill, then you can go for those roles. Singing lessons are fantastic for helping you understand your instrument. I got my masters and bachelor's in voice performance and lemme tell ya, it really made a difference when making the switch to VO. VO coaching will give you acting and delivery tips, help you figure out your niches, and be a great professional resource for career advice.
Huge congrats on going for it!! I highly recommend learning as much as you can about what makes clean audio, how to do basic cleanup, and mic technique. A little bit every day, and you're gonna shock yourself with how much you improve.
